The In-Isa Shrine in Zelda: Tears of the Kingdom is the second shrine Link must complete on the Great Sky Island. Here is a full walkthrough on how to complete it and get the ability to Fuse items together.
Zelda: Tears of the Kingdom begins on the Great Sky Island. From there, you have to complete four shrines and unlock Link's new abilities. The second shrine in line is called "In-Isa". It is here that you will learn how to use the Fusing ability. In this walkthrough, we will guide you through the In-Isa Shrine to complete the “Ability to Combine” trial and receive a Light of Blessing in the end.

Zelda: Tears of the Kingdom In-Isa Shrine Location
After getting the Ultrahand ability and completing the Ukouh Shrine, your next target will be the In-Isa shrine. It is located on the south-west side of the Great Sky Island. Here is the exact shrine location:

After entering the In-Isa Shrine, Rauru will give you the Fuse ability. With this, you'll have the power to attach items to an equipped weapon or shield to enhance it.

Room 1
Now move ahead inside the shrine. You'll find a Rusty Claymore, some boulders and a large stone wall that is blocking the entrance to the next room. In order to destroy the stone wall, you need to create a weapon with more power. To do that, you must equip the Rusty Claymore and use fuse to combine it with one of the boulders in the room to create a Boulder-Hammer.
With the boulder attached to your Claymore, you can now hit the object, that is blocking the entrance to the next room.
Room 2
The second room contains four stone pillars in each corner. Hit the left pillar in the back to get a chest that contains 5 arrows. To your right, you'll find a room with a bunch of plants that carry "Fire Fruits". Collect everything you can find, including the bow and arrows on the ground.
If you take a look up in the room, you will find a box that is attached to the wall with a wooden panel. You can bring it down by shooting it with burning arrows. This is where the fire fruits come into play.
Draw your bow by pushing down ZR, press and hold the upward button on your D-pad and select the item you wish to Fuse to your arrows (in this case: Fire Fruit).
Now you can shoot down the chest, and collect the small key that it contains. Use it to open the door from the previous room.
Room 3
Before you can head to the end of the shrine and grab the Light of Blessing, you have to defeat a Zonai construct. This is pretty easy if you just shoot the vines on the ground with your fire fruit fused arrows.
After smackin' that Zonai construct's butt, just smash the stone objects like you did in the beginning. Et voilà! There you have it, all you have to do now is run up to the altar and grab your Light of Blessing.
